SWATH Technology
SWATH (Sequential Window Acquisition of all Theoretical fragment ions) is an innovative mass spectrometry acquisition mode, classified as a DIA (Data Independent Acquisition) technology. SWATH divides the entire mass spectrometry scan range into several small windows, sequentially fragmenting all ions in each window, allowing for high-speed primary MS scanning of all peptide ions within the scan range and further secondary MS/MS analysis of all precursor ion fragments. SWATH technology can identify all molecules in complex samples, including low-expression proteins and low-abundance modified proteins, characterized by high throughput and high precision.
SWATH Proteomics
SWATH proteomics primarily utilizes SWATH technology for quantitative proteomic research. Similar to the label-free technique, SWATH can quantify sample proteins without labeling. The SWATH quantitative proteomics process involves protein extraction and purification from samples like cells or tissues, followed by reduction to break protein disulfide bonds and enzymatic digestion into peptide fragments using enzymes like trypsin. The peptide mixture is then separated and analyzed using LC-MS chromatography-mass spectrometry tandem analysis, followed by SWATH mass spectrometry analysis. Finally, software analysis of the mass spectrometry data allows for protein identification and quantification. SWATH requires high instrument scanning speed and detection accuracy, with commonly used platforms for SWATH analysis being the AB SCIEX Triple-TOF 5600-plus or higher-end versions of similar mass spectrometers.
