Peptide Synthesis - What is the purity of crude peptides and desalted peptides? How to purify peptides? What are impurities?
For short peptides with a normal sequence of 15 amino acids or less, the HPLC concentration for crude grade is generally 40-60%; for desalted grade, the HPLC concentration is generally 50-70%. The longer the peptide, the lower the purity of the crude or desalted product. Peptides are typically purified using HPLC with a water and acetonitrile gradient. Most impurities are fragments or missing peptides, incompletely deprotected peptides, and residual salts and water.
