Peptide Synthesis - How Consistent Are Research-Grade Custom Peptides Between Batches?
Quality control of research-grade custom peptides is limited (only MS and HPLC analysis). Therefore, differences between batches in terms of desired peptide content, salt content, water content, impurities, solubility, etc., are expected. However, these batch-to-batch differences will not affect most experiments. If you are engaged in sensitive bioassays and wish to minimize batch-to-batch variability, you should request additional quality control and analysis (e.g., peptide content, salt content, water content, solubility). For instance, if you are involved in cell-based assays, you should request the removal of TFA (<1%); if you need to apply the same peptide concentration in experiments across different batches, peptide content analysis is required; if you require high solubility or concentration in specific solvents between batches (e.g., 5mg/ml), you should request solubility control and testing. To achieve ultimate batch-to-batch consistency, you may need to consider GMP-grade peptides.
