MS-MS Measurement of Protein Content and Purity
MS-MS involves selecting meaningful peptide ions for further secondary mass spectrometry analysis based on primary mass spectrometry. This means performing two consecutive mass spectrometry analyses on the sample to be tested, usually accomplished through tandem mass spectrometers such as Triple Quadrupole Mass Spectrometer (TQMS or QqQ), Hybrid Triple Mass Spectrometer Quadrupole Trap MS, and Hybrid Linear Ion Trap Orbitrap MS.
Mass spectrometry technology primarily identifies the relative molecular mass of proteins. By combining it with other techniques such as chromatography, it can characterize additional physicochemical properties, including protein amino acid sequence, content, concentration, purity, post-translational modifications, and protein-protein interactions. High-Performance Liquid Chromatography-Tandem Mass Spectrometry (HPLC-MS-MS) is an important mass spectrometry technology for analyzing protein content and purity. MS-MS couples two mass spectrometers, adding a collision reaction cell between the two quadrupoles to fragment precursor/parent ions selected by the first quadrupole, generating fragment/daughter ions. The secondary mass spectrometry further fragments ions broken down by the primary mass spectrometry, allowing for more detailed sample information with good reproducibility, resolution, and sensitivity, improving the accuracy of detection results.
Biotech company Biotai Parker utilizes Thermo Fisher's Q ExactiveHF mass spectrometry platform, combined with Nano-LC chromatography, to provide efficient and rapidprotein mass spectrometry analysisservice packages.
