HDX-MS

Hydrogen deuterium exchange mass spectrometry (HDX-MS) is a novel mass spectrometry-based technique for studying the spatial structure of proteins. Its basic principle is that the hydrogen atoms on the surface and inside the protein exchange with deuterium atoms in heavy water at different rates. The hydrogen atoms on the surface, being in close contact with heavy water, exchange more readily with deuterium atoms. Through mass spectrometry detection, the different exchange rates corresponding to proteins and peptide fragments can be determined, thereby inferring the spatial structure of the protein.

Hydrogen deuterium exchange mass spectrometry can quickly identify amino acids located on the protein surface and has wide applications in the study of protein interaction sites, epitope activity sites, and dynamic changes of epitopes.
