MALDI TOF Identification of Peptides

Mass spectrometry is a powerful analytical technique commonly used in proteomics research and holds an important position in the characterization of proteins/peptides. It enables qualitative and quantitative identification, molecular weight determination, molecular structure analysis, amino acid composition and sequence analysis, as well as the identification of post-translational modifications of proteins/peptides. Mass spectrometry achieves these analyses by ionizing the protein or peptide samples into charged ions and measuring their mass-to-charge ratio (m/z).

The direct research object of mass spectrometry is ions, so samples must first be ionized into ions using an ion source. Matrix-Assisted Laser Desorption/Ionization Time-of-Flight Mass Spectrometry (MALDI TOF) identifies peptides by using matrix-assisted laser desorption/ionization, which ionizes peptide samples into uniform peptide ions through a soft ionization method. These peptide ions are then detected for their mass-to-charge ratio (m/z) using a Time-of-Flight (TOF) detector. The obtained mass spectrometry data is compared with theoretical data in databases and bioinformatics analysis is conducted to calculate the relative molecular mass information of each peptide ion and deduce the relative molecular mass information of the complete peptide.
