Peptide Purity Analysis
Peptide purity refers to the amount of the target peptide relative to all analytes, i.e., the proportion of the target peptide compared to impurities. Peptide purity can be determined by HPLC analysis, typically detected at a wavelength of 220nm, where peptide bonds absorb maximally. Peptide purity is a key factor affecting the results of a research project. Common contaminants can arise during chemical synthesis, from culture media, or during extraction and purification. Impurities consist of peptides with missing sequences, truncated sequences, incompletely deprotected peptides, and by-products generated during synthesis or final cleavage.

Reverse Phase HPLC
The chromatogram generated by reverse phase HPLC can be used to determine peptide purity. The purity of the peptide is quantified by the peak area of the final purified peptide, which corresponds to the main peak area relative to the total area of all detected peaks in the chromatogram. By calculating the peak area, we can obtain the quantity and relative amounts of by-products.
