HPLC Peptide Map Analysis

HPLC Peptide Mapping Analysisis a routine identification test,primarily used for analyzing and identifying peptide fragments generated from protein enzymatic digestion.The basic principle is to enzymatically digest protein samples into multiple peptide segments, which are then separated byreversed-phasehigh-performance liquid chromatography (RP-HPLC).After separation through the chromatography column, peptides are detected using a UV detector at a wavelength range of 200-230nm (typically 214nm), and a chromatogram is recorded.Finally,the chromatogram of the test sample is compared to that of a reference standard, achievingaccurate identification and quantificationof the target peptide segments,ultimately used in product release tests for identification, assessing inter-batch consistency of the production process, and stability of cell matrix expression used in production.
