Untargeted Proteomics Analysis
Untargeted proteomics analysis, also known as exploratory proteomics analysis, is a method for comprehensively analyzing all detectable proteins in a biological sample without pre-selecting specific proteins as study targets. The goal of this approach is to identify and quantify as many proteins in the sample as possible, including those that are unknown or unexpected, to reveal the functional state of cells, disease mechanisms, or biomarkers.
I. Main Characteristics
Comprehensiveness: Untargeted proteomics aims to comprehensively analyze the protein composition in samples, including both high-abundance and low-abundance proteins.
Unbiasedness: Since there is no need to pre-select target proteins, this method can unbiasedly discover new or unexpected proteins and functions.
Technology Dependence: High-resolution mass spectrometry techniques (such as LC-MS/MS) are key tools for conducting untargeted proteomics analysis.
II. Analysis Workflow
Sample Preparation: Includes protein extraction, purification, and digestion, breaking down proteins into peptides that can be analyzed by mass spectrometry.
Mass Spectrometry Analysis: Using liquid chromatography-mass spectrometry (LC-MS/MS) to analyze peptides, generating a large amount of mass spectrometry data.
Data Processing and Analysis: Using bioinformatics tools to process mass spectrometry data, including peptide identification and quantification, and protein inference.
Biological Interpretation: Classifying and interpreting detected proteins through database comparison and functional annotation to uncover their possible biological significance.
III. Application Areas
Disease Mechanism Research: Comparing the proteome of healthy and diseased states to find disease-related proteins and pathways.
Drug Target Discovery: Identifying proteins or pathways affected by drugs to provide targets for new drug development.
Biomarker Development: Discovering protein markers that can be used for disease diagnosis, prognosis assessment, or treatment monitoring.
Systems Biology Research: Constructing protein interaction networks to understand complex biological processes and signaling pathways within cells.
