Mass Spectrometry Detection Protein Quantification Process

1. Sample Preparation:

2. Protein Digestion:

Enzymatic Digestion: Treat protein samples with enzymes (commonly trypsin) to break down the proteins into smaller peptides.

3. Quantitative Labeling of Peptides (Optional):

Labeled Quantification: Techniques such as Isobaric Tags for Relative and Absolute Quantitation (iTRAQ) or Stable Isotope Labeling with Amino acids in Cell culture (SILAC) use chemical or isotope labeling for peptide quantification.

Label-free Quantification: Directly perform label-free quantitative analysis without using labels.

4. Liquid Chromatography (LC) Separation:

Peptide Separation: Use liquid chromatography (LC) to separate peptides, reducing sample complexity and enhancing the efficiency and sensitivity of mass spectrometry detection.

5. Mass Spectrometry (MS) Analysis:

MS Detection: The peptides separated by LC are sent to a mass spectrometer for detection, where identification and quantification are performed based on the mass-to-charge ratio (m/z) and fragmentation patterns of the peptides.

6. Data Analysis:

Protein Identification and Quantification: Use specialized software and databases to process mass spectrometry data, identifying peptides and proteins and performing quantitative analysis.

Bioinformatics Analysis: Further analyze protein functions, interaction networks, and biological pathways.

7. Result Validation (Optional):

Independent Validation: Validate mass spectrometry results using techniques such as Western blotting or enzyme-linked immunosorbent assay (ELISA).
