Quantitative Acetylation Proteomics Study

Acetylation modification is a highly conserved reversible protein modification in the body, playing a crucial role in the activation of transcriptional regulators within the cell nucleus. Additionally, a large number of non-histone acetylation modifications are involved in the regulation of metabolic pathways and metabolic enzyme activities.

Due to the low abundance and wide dynamic range of acetylated proteins in biological samples, enrichment of acetylated peptides is required prior to mass spectrometry analysis to enhance their abundance. Traditional quantitative proteomic analysis techniques are then used to quantify the enriched acetylated peptides. BTP Company utilizes peptide pre-separation to reduce the impact of high-abundance histones on protein acetylation identification. This is combined with immunoprecipitation using highly efficient antibody enrichment of acetylated peptides, enabling large-scale acetylation identification and quantification. The selected kits are internationally recognized acetylated peptide-specific enrichment kits provided by CST Company. To identify and quantify more acetylated peptides, we use 2-3 different enzymes for protein digestion during the enzymatic digestion process, ensuring that peptides with acetylation sites are efficiently ionized and introduced into the mass spectrometer for analysis, avoiding the loss of acetylation information due to unsuitable peptide length or low ionization efficiency.

Acetylation Quantitative Proteomics Analysis Sample Requirements

1. If you provide tissue samples, please send them to us on dry ice.
2. If you provide protein samples, use ordinary tissue or cell lysis buffers for protein extraction.
3. Sample transportation: Please use sufficient dry ice for transportation and choose faster shipping methods to minimize the risk of sample degradation during transport.
4. Before the formal experiment, we will test the samples you provide. Formal experiments will only begin if the samples pass the test.

Research Cases

This project involves the identification of protein ubiquitination modification sites in Co-IP samples from a teacher at the Institute of Biophysics, Chinese Academy of Sciences, conducted at BTP. Part of the results is shown in the figure below:
