Glycoprotein Structure Analysis
Glycosylation is the process by which a sugar chain is covalently attached to the amino acid side chains of proteins in the form of a glycosidic bond under the catalysis of glycosyltransferases, ultimately forming glycoproteins. Studies have shown that more than 50% of eukaryotic proteins participate in life activities in the form of glycoproteins after undergoing glycosylation, playing their roles. Analyzing glycoprotein structures is an important prerequisite for understanding their physiological functions.
Glycoprotein structural analysis based on mass spectrometry can be conducted using liquid chromatography-tandem mass spectrometry (LC-MS/MS) or hydrophilic interaction chromatography-tandem mass spectrometry (HILIC-MS/MS) techniques. The basic process is as follows: first, the specifically modified glycoproteins are enzymatically cleaved into small glycopeptide fragments using proteases, and the resulting glycopeptides are purified and enriched. Then, LC-MS/MS or HILIC-MS/MS is used for sample measurement to obtain the raw mass spectrometry data. Finally, based on composition and structural fingerprints, site and structural specificity databases, the obtained glycopeptide sequences are searched and analyzed to complete glycoprotein structure analysis and identification.
