Correct Methods for Analyzing Post-Translational Modifications of Proteins

Post-translational modification (PTM) refers to the process by which the biochemical properties of proteins are further altered after translation. These changes may include phosphorylation, acetylation, glycosylation, ubiquitination, etc. Understanding the process of post-translational modification is significant for decoding life information, deepening the understanding of biological principles, and studying the mechanisms of disease occurrence and development.

1. Protein Mass Spectrometry

Protein mass spectrometry is currently the mainstream method for studying post-translational modifications. It mainly includes liquid chromatography-tandem mass spectrometry (LC-MS/MS) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). These methods allow us to directly detect the presence and location of post-translational modifications. Additionally, mass spectrometry can be used for quantitative analysis of the extent or distribution of post-translational modifications.

2. Bioinformatics Methods

With the development of bioinformatics, many prediction tools such as GPS and ModPred have been developed to predict potential post-translational modification sites on proteins, providing a theoretical basis for experimental design.

3. Network Biology Methods

Through network biology methods, protein-protein interaction networks (PPI) can be constructed to further infer the possible functions and biological significance of post-translational modifications. For instance, software like STRING and Cytoscape can be used to construct and analyze PPI networks.

4. Experimental Validation Methods

Although mass spectrometry and bioinformatics methods provide us with a wealth of information, this information needs to be validated by experimental methods. Common experimental validation methods include Western blotting, co-immunoprecipitation (Co-IP), enzyme-linked immunosorbent assay (ELISA), etc. These methods help us verify the existence of post-translational modifications and their effects on protein function.
