Phosphorylation modifications are simple, flexible, and reversible, with the accessibility of ATP as a phosphate group donor, making it a common regulatory mechanism in eukaryotic cells. The reversible process of protein phosphorylation and dephosphorylation regulates nearly all life activities, including cell proliferation, development, differentiation, cytoskeleton regulation, apoptosis, neural activity, muscle contraction, metabolism, and tumorigenesis.
Due to the low abundance and wide dynamic range of phosphorylated proteins in biological samples, quantitative proteomics analysis of enriched phosphorylated peptide samples requires enrichment to increase the abundance of modified proteins. Based on experimental needs, BiotechPack selects solid-phase metal affinity chromatography (IMAC), phospho-specific antibody enrichment, or TiO2 enrichment to achieve efficient sample enrichment. During the peptide enrichment process, mono-phosphorylated and multi-phosphorylated peptides are stepwise separated for mass spectrometry analysis, avoiding the impact of ionization efficiency issues of multi-phosphorylated peptides on the identification and quantification of mono-phosphorylated peptides during mass spectrometry analysis.

BTP-Phosphorylation Quantitative Proteomics Sample Requirements
1. If you are providing tissue samples, please send them on dry ice; plant tissue samples should be over 200mg, blood samples at least 1ml (ensure plasma uses EDTA anticoagulant), serum 0.5ml, urine 2ml, animal tissue samples at least 1g, cell samples should be 5X10^7 cells, and yeast or microbial samples should have a dry weight of 200mg.
2. If you are providing protein samples, please ensure the total protein amount is above 1mg; regular tissue or cell lysis buffers can be used for protein extraction.
3. Sample transportation: Please use sufficient dry ice for shipping and choose the fastest possible delivery method to minimize the risk of sample degradation during transit.
4. Before the formal experiment, we will test the sample you provide, and the official experiment will begin once the sample passes the quality check.
