Multi-pathway Phosphorylation Proteomics

Protein phosphorylation is a reversible post-translational modification in which the amino acid residues of proteins are phosphorylated by protein kinases through the addition of covalently bound phosphate groups. Phosphorylation modification alters the structural conformation of proteins, leading to their activation, inactivation, or functional change. Studying protein phosphorylation helps in understanding biological activities and elucidating the mechanisms of diseases related to abnormal protein phosphorylation. Due to the low abundance and wide dynamic range of phosphorylated proteins in biological samples, it is necessary to enrich phosphorylated peptides before quantitative analysis of phosphoprotein samples using quantitative proteomics methods to increase the abundance of modified proteins.

Bio-Techne utilizes CST's Multi-Pathway Enrichment Kit for enrichment, combined with Thermo Fisher's Orbitrap Fusion Lumos mass spectrometry platform and nanoLC-MS/MS nanoscale chromatography, to provide a multi-pathway phosphoproteomics analysis service package. Simply inform us of your experimental objectives and send your samples. Bio-Techne will handle all subsequent project tasks, including protein extraction, proteolysis, phosphorylation peptide enrichment, peptide separation, mass spectrometry analysis, raw mass spectrometry data analysis, and bioinformatics analysis.

About the samples

1. If you are providing tissue samples, please send them on dry ice; plant tissue samples should be over 200mg, blood samples at least 1ml (use EDTA for plasma anticoagulation), serum 0.5ml, urine 2ml, animal tissue samples at least 1g, cell samples should be 5x10^7 cells, and yeast or microorganisms should have a dry weight of 200mg.
2. If you are providing protein samples, ensure that the total protein quantity is above 1mg; ordinary tissue or cell lysis buffer can be used during protein extraction.
3. Sample transportation: Please use sufficient dry ice for transportation and choose a faster postal method to minimize the possibility of sample degradation during transit.
4. Before the official experiment, we will conduct a test on the samples you provide, and the formal experiment will begin once the samples pass this test.
