2D-DIGE Antibody Coverage Analysis

Two-dimensional fluorescence difference gel electrophoresis (2D-DIGE), also known as two-dimensional fluorescence differential gel electrophoresis, is a proteomics technique used for qualitative and quantitative comparison of protein expression changes in different samples (such as diseased and healthy tissues). It plays a crucial role in the production of biological products, especially in HCP antibody coverage analysis. HCP antibody coverage analysis is typically used to assess the sensitivity and specificity of antibodies for detecting HCP. The 'coverage' here usually refers to the proportion of HCP types that can be detected by the antibody out of all HCP types. A high-coverage antibody means it can detect more types of HCP, thus providing a more comprehensive assessment of impurity levels in the production process.

 

In 2D-DIGE antibody coverage analysis, proteins from different samples are labeled with different fluorescent dyes and then mixed for two-dimensional gel electrophoresis separation. By marking the HCP protein spots on the two-dimensional electrophoresis map that react with the antibody, the specificity and coverage of the antibody can be determined. Combined with mass spectrometry, the specific types of HCP covered by the antibody and important HCP that may be missed can be accurately identified. The 2D-DIGE technique offers higher resolution, reproducibility, and accuracy compared to traditional two-dimensional electrophoresis, and has been widely used to compare HCP expression levels under different production batches or conditions to evaluate process consistency and control.
