Acetylated Protein WB
Acetylation modification is a reversible and highly conserved post-translational modification of proteins, often occurring in histones and non-histones. It plays an important regulatory role in gene expression, transcription factor activity, cellular metabolic pathways, and metabolic enzyme activity. WB (Western blotting), also known as immunoblotting or protein blotting, is an immunochemical technique developed on the principle of specific antigen-antibody binding, combining gel electrophoresis and immunochemical techniques to detect a specific protein in complex samples, allowing for qualitative and semi-quantitative analysis.
Acetylated protein WB uses immunoblotting technology to detect acetylated proteins. The general experimental procedure involves transferring protein samples separated by PAGE to a solid-phase carrier. The proteins on the solid-phase carrier act as antigens that immunologically bind to the corresponding antibody (primary antibody), then specifically bind to a specially labeled secondary antibody. Finally, detection is performed using substrate coloration or autoradiography techniques, enabling qualitative identification of acetylated proteins.
