Is DDA a non-labeling quantitative proteomics method?
DDA (Data-Dependent Acquisition) is one of the label-free quantitative proteomics methods. In proteomics research, quantitative methods are generally divided into two categories: labeled quantification and label-free quantification. DDA is a commonly used label-free quantification method that relies on the data-dependent mode of mass spectrometers to select specific precursor ions for further fragmentation analysis. This method does not require prior chemical labeling of proteins or peptides, thereby simplifying the sample processing procedure. DDA is widely used in proteomics, especially in the identification and quantitative analysis of complex samples.

The key points of DDA (Data-Dependent Acquisition) technology are as follows:
1. Basics of Mass Spectrometry:
DDA is a label-free quantitative proteomics method based on mass spectrometry technology. It identifies proteins by analyzing the mass and fragmentation spectrum of peptides or proteins.
2. Automatic Selection of Precursor Ions:
During the MS (mass spectrometry) scan, DDA automatically selects a certain number of peptide ions (precursor ions) with the strongest signals for MS/MS (tandem mass spectrometry) scanning.
3. Fragmentation Analysis:
The selected peptide ions are fragmented during MS/MS scanning, producing specific fragmentation patterns. These patterns are crucial for identifying specific protein sequences.
4. Application Range:
DDA is suitable for protein identification and quantitative analysis in complex samples and is widely used in proteomics research.
5. Advantages:
Easy to operate, applicable to a wide range of sample types, and capable of effectively identifying and quantifying various proteins.
6. Limitations:
Sensitivity to detect low-abundance proteins is relatively low, and the reproducibility of results is influenced by sample complexity and mass spectrometer performance.
