Acetylation Quantitative Proteomics Research

Acylation modification is a type of post-translational modification of proteins. Important cellular metabolites, acyl-CoA compounds, act as donors directly participating in protein acylation modification in organisms, thereby regulating various important biological processes such as epigenetics, energy metabolism, protein transport, and interactions. Protein acylation modification is currently one of the hotspots in life science research. Among protein acylation modifications, acetylation was the first to be discovered and studied extensively. In recent years, other types of acylation modifications such as propionylation, malonylation, succinylation, crotonylation, and others have been discovered. Quantitative proteomics studies of acylation modifications can provide deep insights into the close relationship between changes in protein acylation modifications and life activities by comparing the levels of acylation modifications in biological samples under different physiological or pathological states. This is of great significance for revealing the mechanisms of life activities, screening clinical biomarkers for diseases, identifying drug targets, and more.

In quantitative proteomics analysis of acylation, protein samples are first digested into peptide mixtures. Then, liquid chromatography is used to separate the components of the digested peptide mixtures to reduce sample complexity. Some types of acylation-modified peptides can be specifically enriched using high-quality acylation modification antibodies or kits, and finally analyzed quantitatively through LC-MS/MS.
