Hello, is there a methylation NMR analysis method?

Protein methylation is a common post-translational modification that has a significant impact on regulating protein functions, stability, and interactions. NMR can provide detailed information on the identification of methylation sites, methylation dynamics, and methylation status of proteins. Utilizing NMR technology for analysisThe general steps for protein methylation are as follows:

1. Sample Preparation:

Select appropriate methyltransferases and donors, such as SAM (S-adenosyl methionine), to methylate the target protein.

2. Labeling Strategy:

By selectively labeling methylation sites on proteins, the sensitivity and resolution of NMR can be enhanced. For example, specific isotope labeling (such as 13C, 15N) can be used to facilitate the identification of methylation sites.

3. NMR Data Collection:

Use conventional one-dimensional or two-dimensional NMR experiments, such as HSQC or TOCSY, to collect data.

4. Data Analysis:

By analyzing NMR peaks, the chemical environment of methylation sites can be determined. For proteins with known structures, the specific methylation sites can be identified by comparing NMR data with those of non-methylated proteins.

5. Structural Modeling:

For proteins with unknown structures, NMR data and other structural information can be used to construct a 3D structural model of the methylated protein.
