DIA Proteomics Sample Processing Steps
Data independent acquisition (DIA) proteomics uses DIA technology (such as SWATH technology) to detect and analyze the proteome in samples. Biotyper Biotech provides mass spectrometry-based DIA proteomics analysis services and protein sample preparation services.
DIA Proteomics
DIA is a holistic mass spectrometry data acquisition mode. After primary mass spectrometry detection, DIA fragments all parent ions within a specific mass-to-charge ratio range, collects the fragment ions of all parent ions, and quickly sequentially scans all fragment ions within adjacent wide windows of parent ions, thereby obtaining complete peptide information and greatly improving data reproducibility. DIA proteomics can perform relative or absolute quantification of the proteome.

DIA Proteomics Sample Processing Steps
Due to the complexity of the proteome, there is no standard method for preparing protein samples for mass spectrometry (MS) analysis. The specific sample processing scheme depends on the sample type, experimental goal, and analytical method used. When designing a sample processing strategy, many factors need to be considered, including the source, type, physical characteristics, abundance, complexity, and cellular location of proteins. Like other mass spectrometry-based proteomics, DIA proteomics generally includes sample processing steps such as cell lysis, high-abundance protein depletion or target protein enrichment, and protein reduction and enzymatic digestion.
