What mass spectrometry methods are used for peptide identification

Peptide identification can be performed through peptide fingerprinting or tandem mass spectrometry. The method involves enzymatically cleaving peptide segments into small peptide mixtures using proteases, followed by mass spectrometry analysis to obtain the amino acid sequences of the small peptide segments for identification. Peptide identification requires two levels of mass spectrometry analysis. The peptide mixture is subjected to primary mass spectrometry to select high-abundance peptide molecules for secondary mass spectrometry. In secondary mass spectrometry, peptide segments collide with each other causing amino acid bond breakage. The resulting peptide fragment ion signals are collected by the detector and a secondary mass spectrum based on mass-to-charge ratio (m/z) is outputted. By interpreting the spectrum, the amino acid sequence of the peptide segment can be analyzed and identified.
