TMT Quantitative Proteomics Analysis
TMT technology is a chemical labeling technique developed by Thermo, USA. It uses isotopic tags to specifically label the N-terminus or lysine side chain groups of peptides obtained after enzymatic digestion, followed by mass spectrometry for relative quantification of proteins. TMT quantitative proteomics analysis refers to the use of TMT technology to perform relative quantitative analysis of all proteins in cells, tissues, or organisms. It is a mass spectrometry-based technology for determining the relative protein content and differential protein expression analysis in different samples. TMT quantitative proteomics analysis mainly involves steps such as trypsin digestion, TMT labeling of peptides, liquid chromatography separation, mass spectrometry detection, and bioinformatics analysis.
Compared to label-free proteomics analysis, TMT quantitative proteomics analysis is limited by the number of tags and can currently detect up to 16 groups of samples simultaneously. TMT-based quantitative proteomics analysis allows for simultaneous on-machine detection of mixed samples, reducing individual detection errors to a certain extent. It offers higher detection throughput and is advantageous for detecting low-abundance proteins.
