Differences between SRM, MRM, and PRM

MRM (Multiple Reaction Monitoring), also known as SRM (Selected Reaction Monitoring), and PRM (Parallel Reaction Monitoring) are two different mass spectrometry data acquisition modes used in targeted quantitative proteomics research. PRM was developed based on MRM/SRM, and the main differences between the two are as follows:

(1) MRM/SRM monitors the transition of each precursor ion/product ion individually, while PRM analyzes all product ions from a precursor ion with high resolution and high mass accuracy;

(2) PRM has mass accuracy up to ppm level, which can better eliminate background interference and false positives compared to SRM/MRM, effectively improving detection limits and sensitivity in complex backgrounds;

(3) PRM not only has quantitative analysis capabilities but also qualitative capabilities;

(4) PRM does not require the selection of ion pairs and optimization of fragmentation energy, making it easier to establish analyses;

(5) PRM is usually conducted on high-resolution hybrid quadrupole-orbitrap (Q-OT) or time-of-flight instruments, while MRM/SRM is based on triple quadrupole mass spectrometers.
