Protein purification is a fundamental experimental technique used to extract and purify the protein of interest and obtain a sufficient quantity of sufficiently pure protein samples. The following are some common protein purification methods:
1. Precipitation Method:
This method mainly utilizes the solubility differences of proteins under specific pH, electrolyte concentration, temperature, organic solvent, and other conditions for precipitation separation. Common protein precipitants include ammonium sulfate, acetic acid, PEG, etc.
2. Chromatography:
Chromatography is a powerful tool for finely separating molecules according to their biochemical properties. It mainly includes affinity chromatography, ion exchange chromatography, gel filtration chromatography, reverse-phase chromatography, etc.
3. Electrophoresis:
Proteins with different surface charges migrate at different rates in an electric field, and during electrophoresis, they can be separated based on the differences in the distances they migrate in the gel. Common electrophoresis methods include agarose gel electrophoresis and polyacrylamide gel electrophoresis.
4. Ultracentrifugation:
This method uses the mass differences of different proteins. Under the action of centrifugal force, heavier proteins or components will precipitate faster, while lighter ones will suspend or move more slowly, thereby separating different proteins. It mainly includes density gradient centrifugation.
The above methods usually need to be used in combination to obtain high-purity protein, and specific purification steps need to be adjusted according to the properties of the target protein.
